Hong-Ou-Mandel (HOM) interference between two indistinguishable photons is a cornerstone of photonic quantum information science and underlies Bell-state measurement (BSM) in measurement-device-independent quantum key distribution (MDI-QKD). Although HOM interference is often associated with photon bunching that requires simultaneous arrival of two photons, its physical origin lies in the indistinguishability of two-photon probability amplitudes, which does not fundamentally require temporal overlap. Building on this understanding, we experimentally demonstrate HOM interference between temporally non-overlapping optical pulses and study the coherence conditions. Extending this to Bell-state measurement, we propose an asynchronous two-mode encoded MDI-QKD protocol that achieves a loss-to-key-rate scaling comparable to twin-field and mode-pairing QKD. We also report preliminary experimental results supporting this approach.
